 >25 years of experience working with Indian and global academic organizations and pharmaceutical industries. Led and managed the discovery portfolio of discovery programs on complex human diseases in the area of metabolic and immunological disorders (Diabetes, Obesity, Metabolic associated- Steatotic Liver Disease (MASLD), cancer, respiratory and autoimmune diseases) using small molecule First-in-Class Targeted approaches and herbal natural products while working with inter-disciplinary institutional and global teams in leading pharmaceutical companies during 2002-2017.

Current Interests: Metabolic-associated Steatotic Liver Disease (MASLD)

Metabolic-associated Steatotic Liver Disease (MASLD) is a slow-progressing disease with aberrant lipid metabolism, inflammation, oxidative stress, and fibrosis which has nearly 30 % global prevalence with limited treatment options available to date. Current focus of my lab is to understand the pathophysiology and identify potential therapeutic options for the management of MASLD and associated comorbidities.

My lab has established in vitro and in vivo models to perform pre-clinical assesment of small chemical entities as well as natural product formulations to identify potential therapeutic options for the management of NAFLD. 

Apart from conventional 2D models my lab has established multilineage 3D spheroid and human liver organoid models mimicking the liver heterogeneity to test the anti-steatotic, anti-inflammatory and anti-fibrotic properties of test compounds.

Establishment of In Vitro Assays

  • Established Cell-free assays for the screening of compounds against the targets related to diverse molecular classes such as Kinases, GPCRs, Nuclear Receptors, phosphatasess, other enzymes and biochemical and signaling pathways.
  • Established 2D and 3D spheroid cell based assays using monoculture and co-culture technology to mimic the key hallmarks of MASLD such as steatosis, inflammation and fibrosis.

Establishment of Human Liver Organoid Platform

We are one of the very few research labs in India to establish a human tissue-derived organoid platform to understand the pathophysiology and conduct drug discovery research and tissue regeneration studies on MASLD using liver tissues from healthy humans and MASLD patients.  We have adopted a hepatocyte progenitor cell approach to convert resident primary hepatocytes and biliary cells into organoids with bipotent and stem cell functions.  Our lab has established a steatohepatitis model using organoids to mimic the key phenotypes of MASLD. This model is currently being employed to develop therapeutics, tissue regeneration, and identify novel biomarkers for diagnostic and prognostic purposes as per the following specificatios:

-As a screening model for the development of Therapeutics (Findings published in J Cell Exp Hepatol, 2025)

-As a cell therapy approach for chronic liver disease  (A Bio-E3 Concept)

- For the development of Precision Medicines for MASLD and HCC

- Identification of  stage-specific molecular signatures of MASLD for the development of diagnostics

 

Establishment of Diet-induced Mice Models of MASLD

My laboratory has established diet-induced mouse models of MASLD using C57/BL-6 mice fed with various obesogenic and nutrient-deficient diets followed by analyzing several metabolic, biochemical parameters, and gene expression patterns in metabolic tissues to assess the efficacy of test compounds. We have observed diet-specific modulation of components of innate immunity 

Identification of Small Molecule LEADS as Potential Therapeutic Candidates to manage MASLD

  • A small molecule dietary metabolite has been identified in my laboratory for its ability to modulate the key hallmarks of MASLD using in vitro cell-free and cell-based and mouse models of MASLD. ((Ongoing Studies, Provisional patent filed). 
  • Identification of LEADS for the treatment of MASLD using the drug repurposing approach, from a commercially available library of over 3000 compounds  (Ongoing Studies).

Identification of Herbal Botanical Extracts as Therapeutics to Manage Respiratory Diseases and MASL

 My lab has adopted evidence-based approaches using defined in vitro and in vivo pharmacological setups to demonstrate the translational potential of traditional medicines, and identified the following leads:

-  Detailed pharmacological characterization of AYUSH-64, a poly-herbal formulation (developed by the Central Council for Research in Ayurvedic Sciences (CCRAS), Ministry of AYUSH, Govt. of India) was conducted in my laboratory and it was found to possess potent anti-inflammatory and immunomodulatory properties. This formulation significantly reduced inflammatory cytokines in the in vitro cell-based and mice models of LPS-induced mice models of lung inflammation (Pharmacological Research-Natural Products, 2025).

- Aqueous root extract of Withania somnifera was also found to possess anti-inflammatory properties using cell-based assays and mice models of lung inflammation (Frontiers in Pharmacol, 2023).

- My laboratory has also identified botanical extracts that showed amelioration of the key hallmarks of MASLD in a 20-week model of mice fed with a high fat-choline deficient diet as a combination approach (Heliyon 2023) and as a standalone approach.

Delivered a talk on "Leveraging Tumour Organoid Models for Translational Research in Cancer" and received the Certificate of excellence by the  Nobel Laureate by  Prof. James Allison in the International conference '4th World Congress on Translational Cancer Research & Immunotherapy [WCTCRI] 2025" during Feb 10-12, 2025 at Mahatma Gandhi University of Medical Sciences and Technology, Jaipur organized by Prof. Anil Suri, Director of the Center for Cancer Immunotherapy. (Nobel Laureate Prof. James Allison,  received the Nobel Prize in Physiology and Medicine in 2018 for his pioneering discovery of CTLA4 and its inhibition as a potential anti-cancer therapy by inhibition of negative immune regulation.)
